InSilico Medicine (03696.HK): NLRP3 Inhibitor ISM8969/HT-001 Co-Developed with Hengtai Bio Receives NMPA IND Approval for Parkinson's Disease

NewTimeSpace News: InSilico Medicine Cayman TopCo (stock code: 3696) issued a voluntary announcement that its innovative oral NLRP3 inhibitor ISM8969/HT-001, co-developed with Shenzhen Hengtai Biotechnology Co., Ltd. ("Hengtai Bio"), has received Investigational New Drug (IND) approval from China's National Medical Products Administration (NMPA) for the treatment of Parkinson's disease. Following the announcement in June 2026 of the first human dosing in the Phase I study in Australia, this approval marks the formal commencement of parallel clinical evaluations in China and Australia.

According to the announcement, this clinical trial aims to evaluate the safety, tolerability, pharmacokinetic (PK), and pharmacodynamic (PD) characteristics of ISM8969/HT-001 in healthy participants, as well as its preliminary efficacy in Parkinson's disease patients, laying the foundation for subsequent clinical development.

In preclinical evaluation studies, ISM8969/HT-001 demonstrated balanced druggability characteristics, including good in vitro activity and safety, excellent in vivo PK/PD features, and efficacy against inflammation in multiple mouse disease models covering both acute and chronic inflammatory diseases. Compared with other NLRP3 inhibitors in clinical development that have peripheral limitations, ISM8969/HT-001 is capable of penetrating the blood-brain barrier and entering the central nervous system, thus holding potential for treating diseases associated with neuroinflammation.

The announcement noted that ISM8969/HT-001 is an orally available, blood-brain barrier-penetrating NLRP3 inhibitor, empowered by InSilico Medicine's generative AI platform Chemistry42, and has demonstrated excellent efficacy, good safety, and superior blood-brain barrier penetration characteristics in preclinical studies. Since its nomination as a preclinical candidate compound in December 2024, ISM8969/HT-001 has rapidly advanced in clinical development: receiving FDA IND approval in January 2026 and announcing a co-development partnership with Hengtai Bio in the same month; completing first human dosing in the Australian Phase I clinical study in June 2026, achieving the first clinical milestone; and receiving NMPA IND approval in China in July 2026.
